Nucleotide sequence of the glyceraldehyde-3-phosphate dehydrogenase gene from Cryphonectria parasitica.

The gene for glyceraldehyde-3-phosphate dehydrogenase (gpd-l) of Cryphonectria parasitica, the chestnut blight fungus, was isolated by probing a genomic library with a PCR (1) amplified copy of the coding region of the Aspergillus nidulans homologue, gpdA (2). Presented below is the nucleotide sequence for the entire gpd-l coding region, 381 bp of the 5'-flanking region and 557 bp of the 3'-flanking region. The gpd-l coding region was found to contain two introns which correspond to introns 5 and 6 of the gpdA gene. Conserved 5' and 3' splice signals are indicated by asterisks while internally conserved intron sequences are indicated by # # #. A potential CCAAT motif and polyadenylation signal are underlined. The predicted amino acid sequence of gpd-l is 81% identical to that encoded by gpdA. The gpdA promoter has been successfully used for expression of a foreign gene in several filamentous fungi (3). Transformation and expression studies in C. parasitica utilizing the gpd-l promoter and 3'-flanking region are in progress.
